The demand for stable, accurate, and high-performance cell isolation

Cell isolation is a critical step in many research and diagnostic processes (such as immune cell analysis, stem cell isolation, HLA testing, chimerism, etc.). Traditional manual methods (e.g., gradient centrifugation, handheld magnets with magnetic beads, etc.) are often time-consuming and prone to variability between runs. Common challenges include low target cell recovery, insufficient purity, and the risk of errors or contamination from manual handling. A study at Royal Perth Hospital (Australia) showed that automating T and B cell isolation using the RoboSep™ system could double lymphocyte recovery efficiency from the same sample volume compared to manual methods while significantly reducing processing time. Moreover, the automation process reduces errors during sample handling and avoids cross-contamination by using disposable tips. These results emphasize the importance of a stable, automated cell separation system to meet the needs of current research and clinical laboratories.

RoboSep™-S is the second fully automated cell separator generation in the RoboSep™ series. This device has a compact size and comes with an integrated touchscreen, making it easy for users to operate on-site. Below are the key technical features of RoboSep™-S:

Simultaneous processing of 4 samples

RoboSep™-S can process up to 4 samples simultaneously in one run. Each sample can range from 250 µL to 8.5 mL, making it suitable for various sample sources (whole blood, bone marrow fluid, PBMCs, etc.). With the capacity to process four samples in parallel, the device meets the needs of mid-sized sample processing in research and testing labs.

Sequential isolation of multiple cell types

In addition to parallel separation, RoboSep™-S allows sequential isolation of up to 4 different cell types from the same sample (e.g., sequential isolation of T, B, NK, and mono populations from a blood sample). This feature is especially useful for applications that require the isolation of multiple cell populations from a single sample (such as chimerism analysis).

EasySep™ column-free technology

The device uses EasySep™ magnetic beads technology to label and isolate target cells without the need for columns. By not using columns, the isolation process minimizes cell activation and preserves their biological properties. Nano magnetic beads combined with specific antibodies bind to target cells. A magnet then pulls the bead-bound cells out of the mixture, allowing for the collection of a pure and viable cell population for downstream applications.

Intuitive user interface

RoboSep™-S comes with a color touchscreen integrated directly into the device, featuring a user-friendly graphical interface. Users need to select the desired cell isolation protocol (several pre-programmed protocols for different cell types are available), load the sample and reagents into the tray, and press “Run.” Direct handling time is approximately 5 minutes, after which the machine automatically performs the remaining steps. After 25–60 minutes (depending on the protocol type), users can return to collect the purified isolated cells.

Disposable tip system to avoid contamination

All sample aspiration/dispensing and reagent handling are done via disposable pipette tips, eliminating the risk of cross-contamination between samples. Each sample position has its own magnet and separation component, ensuring independence between processing channels. Importantly, RoboSep™-S does not require daily washing or sterilization after use, making maintenance simpler than systems with fixed liquid pathways.

Chemical tracking and data storage

The machine is equipped with a barcode scanner to identify EasySep™ reagents and manage sample information. After each run, the system can generate an End-of-Run report that records operator details, protocol type, reagent lot numbers, etc., facilitating convenient and transparent data retrieval and management.

Thanks to these features, RoboSep™-S is easily placed in a biosafety cabinet or on a lab bench, and multiple devices can be positioned together to increase throughput when needed. This device is particularly suitable for labs that need to isolate cells on a medium scale, requiring flexibility and ease of use.

RoboSep™-16 is a high-capacity version designed to automate large-scale cell isolation processes. It is ideal for testing centers or research institutes with high parallel sample processing needs. Key features of RoboSep™-16 include:

Simultaneous processing of 16 samples

As its name suggests, RoboSep™-16 can process up to 16 samples simultaneously in one run. Each sample can range from approximately 200 µL to 2 mL, making it suitable for smaller sample volumes (e.g., peripheral blood-derived products, enriched PBMC samples, etc.). With 16 sample positions running in parallel, RoboSep™-16 delivers exceptional throughput to meet the high demand in large labs.

Sequential isolation of multiple cell types from multiple samples

The device allows sequential isolation of multiple cell types from different samples simultaneously. Specifically, RoboSep™-16 can sequentially isolate up to 4 types of cells from 4 different samples in one run. This is helpful when multiple samples need to be processed, and each requires the isolation of multiple cell populations. For example, isolating T and B cells from various patients for immune testing.

Software-operated interface

Instead of an integrated touchscreen, RoboSep™-16 comes with a laptop that has installed control software. The intuitive software interface allows users to set up runs similarly to RoboSep™-S, with only about 5 minutes needed to select protocols and load samples. Using a computer control system also enables flexible software updates, data storage on the system, and easy data import/export.

Smart sensor integration

RoboSep™-16 is equipped with electrically conductive tips with sensors that automatically detect the reagent levels of EasySep™ chemicals and buffers during the run. If a reagent bottle is running low, the system will recognize this and automatically switch to a backup bottle without interrupting the separation process. This smart feature ensures smooth operation even when handling large numbers of samples.

Safety and cross-contamination prevention

Similar to RoboSep™-S, the 16 version uses disposable tips and fully automates all sample and reagent handling steps. This reduces technician exposure to biological samples (increasing biosafety) and eliminates the risk of cross-contamination between samples. RoboSep™-16’s robotic pipetting system and automatic magnets ensure that each sample is handled independently, even when running 16 samples simultaneously.

With its high throughput and flexible features, RoboSep™-16 is perfect for HLA testing labs, blood banks, immune research centers, or any facility that needs to isolate a large number of samples daily.
