STEMprep™ is a benchtop system with a flexible design, suitable for a variety of tissue processing needs in laboratories. Its breakthrough design features temperature-controlled sample holding slots, enabling the adjustment of digestion conditions for each sample. The user-friendly touch screen interface simplifies operations. The compact design measures 284x325x400 mm (LxWxH).
STEMprep™ is designed in a modular way, meeting the diverse tissue processing requirements in research. The Primary Unit can simultaneously process up to 4 samples. When throughput needs increase, the system can easily expand to process 8, 12, or more samples by connecting additional Secondary Units.
This modular approach (see Table 1) allows users to invest based on their current capacity needs and expand flexibly in the future by adding units without replacing the entire system.

How to use STEMprep™
STEMprep™ simplifies sample processing with an intuitive touchscreen interface, enabling quick protocol selection and setup.
Basic operation steps:
- Start the system
Turn the switch at the back of the unit on. The touch screen on the device will display, and the identification labels will appear on all connected units. - Select slot and program
On the Run Control Screen, you will see all available slots. Choose a slot and select the desired protocol from the displayed list. - Prepare the sample
Add the tissue sample along with the STEMprep™ Tissue Dissociation Kit into the STEMprep™ Sample Tube. - Complete the process
Once the protocol is complete, remove the sample tube. The sample is now processed into a single-cell suspension, ready for further analytical applications. The device can be powered off using the switch at the back.
System performance of STEMprep™
Stable performance – Cell viability preserved – Reliable results
Standardises tissue dissociation across different tissue types by minimizing errors from manual handling. By integrating STEMprep™ into your workflow, you can reduce operational errors, automate complex steps, and enhance reproducibility of results.
The cold-hold function preserves tissue sample viability at 4°C, providing flexibility for post-processing analysis, especially useful for heat-sensitive assays such as RNA analysis.
Various mouse tissues processed using the STEMprep™ system in conjunction with the STEMprep™ Tissue Dissociation Kit generate single-cell suspensions. Cell viability and yield have been assessed on multiple tissues, including 4T1 breast cancer (n = 10), B16 melanoma (n = 28), brain (n = 68), liver (n = 37), lung (n = 64), and spleen (n = 35).
Tumors are created by injecting cancer cells subcutaneously into the mouse flank. Tumour, liver, and lung samples are processed with ammonium chloride solution to remove red blood cells. Brain samples are processed with OptiPrep™ 18% (equivalent to 10.8% w/v iodixanol) to remove myelin and cell debris before analysis. After processing with STEMprep™, cell viability and yield are assessed by flow cytometry. Data is presented as mean ± standard deviation (SD).
Mouse lung and spleen tissues are dissociated using the corresponding kits: STEMprep™ Mouse Lung Dissociation Kit and STEMprep™ Mouse Spleen Dissociation Kit. After completing the tissue digestion process, samples are immediately held on the STEMprep™ Tissue Dissociator for up to 24 hours at three different temperatures: 4°C, 22°C, and 37°C. Cell viability of nucleated cells from (A) lung and (B) spleen tissue is measured. Data is presented as mean ± standard deviation (n = 3).
STEMprep™ Sample Tubes
STEMprep™ Sample Tubes are specially designed for use with the STEMprep™ Tissue Dissociator system and compatible tissue processing kits. These sample tubes help users reduce variability, save time, and standardize tissue processing across various tissue types.
Each sample tube incorporates a rotor and cutting blades, activated by the STEMprep device to perform efficient tissue dissociation. This design ensures stable, high-quality results, making it ideal for further analytical applications, such as single-cell analysis or cellular assays.
STEMprep™ Sample Tubes are pre-packaged, ready for immediate use without additional preparation. Place the sample in the tube, seal it, and then insert it into the device to initiate the process. The single-use design helps minimize the risk of cross-contamination, while the durable construction ensures stable operation at the device’s maximum speed.

Tissue-specific enzyme mixtures – Optimise cell viability and consistency
Preparing single-cell suspensions from mouse tissues has never been easier with STEMprep™ Tissue Dissociation Kits. Each kit is designed specifically for a particular tissue type, including brain, liver, lung, spleen, and solid tumours, and is optimized for use with the STEMprep™ Sample Tubes and STEMprep™ Tissue Dissociator.
Tissue processing is carried out gently yet effectively, preserving high cell viability and surface markers, while reducing operator variability and shortening hands-on time. The resulting cell suspension can be immediately used for downstream applications such as cell line isolation, cell culture, flow cytometry, genetic research, or proteomics.
The data shows that these kits provide high recovery efficiency and consistent cell viability across various mouse tissues when used in conjunction with the STEMprep™ system.
