OptiPrep™ is a ready-made, sterile and endotoxin-tested solution of iodixanol use as a density gradient medium recommended for the isolation and purification of DNA or RNA viruses, organelles such as nuclei, mitochondria, endosomes or exosomes, macromolecules, as well as a wide range of cell types. OptiPrep™ is a non-ionic iodixanol-based medium with a density of 1.320 ± 0.001 g/mL. It has been used clinically in X-ray contrast imaging and has been shown to have lower toxicity compared to ionic gradient compounds such as metrizamide.

Storage and Stability Conditions of OptiPrep

Should be stored between 4°C – 30°C

NOTE: Protect product from light. Prolonged exposure to direct sunlight leads to release of iodine from the iodixanol molecule. This effect is negligible when working with this solution on a daily basis.

Before use invert the bottles several times to mix the contents.

Advantages of OptiPrep™ vs Other gradients

OptiPrep™ vs Histopaque & Percoll
Percoll and Histopaque include one or several washing step(s) prior to their use. The removal step(s) is/are required to avoid the formation of density gradients which can be toxic to mammalian cells. Unlike Histopaque and Percoll, OptiPrep™ has no potential for bioaccumulation, therefore is non-toxic for the cell. OptiPrep™ has a better yield, purity and viability compared to Histopaque.

OptiPrep™ vs CsCl (Caesium chloride)
Particle virus number ratios are 100 times higher with OptiPrep™ compared to those recorded from CsCl gradients.
